RATIONAL GENERATION OF DIRECTED PROTEIN-CAPTURE REAGENTS

There are five ovals each with a deep cleft, representing C-clamps, a new class of protein capture reagents that our project aims to develop. The area surrounding the cleft is colored differently in different ovals, schematically indicating that these molecular surfaces are engineered in such a way that they selectively recognize the unique C-terminal signature of their respective target, shown in the matched color. Arrows connecting C-clamps to their respective target represent selective capture by C-clamps. Virtually every protein has a unique C-terminal signature that can be recognized with high efficiency by C-clamps. Thus, C-clamping dramatically simplifies proteome-scale development of protein-capture reagents. The figure also includes a molecular graphics representation of a C-clamp.
